Need to be in Manchester by 7am. Is an hour enough at that time? It's not a commute I've done at that time of day before.

As a rule of thumb us taxi drivers usually allow 75 mins from barnsley to man airport, this is depends on the time of day it could take you as long as 30+ minutes to get past mottram. But travelling at that time you should miss the traffic. I would recommend that you depart barnsley at 05:45 to miss said traffic at mottram.
Been travelling to Stockport fairly regularly with work recently (at peak times) and tend to go via Holmfirth, Saddleworth Moor, Greenfield on the A635- very little traffic in comparison but that bit further north - not sure where you're starting from. Sent from my SM-T710 using Tapatalk
On a normal working week from 6.00am onwards traffic starts to build up through Mottram and is regulated by 2 sets of traffic lights so very slow going. I would normally set off at 5.30am, the traffic tends to be lighter on the Easter week though. So the short answer is God Knows!
I avoid the awful Woodhead pass like the plague and always use the Holmfirth road. It might be worth you doing the same.
I was on a site in Spinningfields, central Manchester a few years ago and was doing it in under an hour leaving at 6am.
I Went that way on Friday. No trouble with traffic. However fog was awful which made it feel very dangerous. As it's such a narrow road

Where in Manchester? Shouldn't be too bad for 7am. Mottram should be ok but might take you 5-10 minutes. Should only be busy on the Mancunian way west bound at that time, unless you need to go on the M60. To get into the centre I'd give yourself just over an hour from Penistone. No need to go through Holmfirth.
